European Union: ?Broke? Cyprus to assume EU presidency

Think of Cyprus and what might first come to mind is the country's enduring feud with Turkey, which has occupied the northern part of the island since 1974.

The country, which joined the European Union in 2004, hopes it can project a different image from July 1, when it takes over the six-monthly rotating EU presidency from Denmark for the first time.

But whether this will be for the better or worse remains open to speculation. Given Cyprus? precarious economic state, Europe watchers are wondering what the country might achieve at the head of a Europe in a similarly parlous position.

?A dog guarding the sausages?

Kurt Lauk, president of the economic council of Angela Merkel's Christian Democrats didn?t mince his words: ?This is the paradox of the European Union, that the dog should be put in charge of the supply of sausages!? He called for all countries that have received bailouts to be barred from holding the presidency.

Heavily exposed to the banking crisis of its big brother Greece, Cyprus is running a debt of 14 billion euros, and its deficit of 6.3 percent is one of the highest in the eurozone.

On Monday it requested a bailout from the EU, which has since been approved. The European Commission, the European Central Bank and the International Monetary Fund (IMF) will begin assessing next week how much bailout money will be needed to prop up the country?s banks.

At the same time, the country?s communist president Demetris Christofias has irked Europe by seeking bilateral aid from Russia and China, albeit at an interest rate 2% higher than from the EU. Cyprus has close ties with Russia, whose economic and political presence in the island has no equivalent elsewhere in Europe.

French daily Le Monde explained the reasoning: ?Cyprus is worried about the possibility of a visit from troika experts that could accompany EU aid. [...] Even more daunting than the possibility of pressure to raise the island?s 10% corporate tax rate, which is the lowest in Europe, Cyprus is alarmed by the prospect of austerity.?

On the ground however, local press report an upbeat mood, with the presidency expected to boost morale about the ailing economy.

?I think the presidency is something that can lift the mood, and maybe distract people?s attention,? said Jacqueline Agathocleous, a journalist for the English daily Cyprus Mail, adding that the arrival of delegates would be good for business.

Turkey in a tizz

The ongoing dispute with Turkey adds another dimension to Cyprus? impending presidency. Turkey occupied the island?s north in 1974, which subsequently declared itself the Turkish Republic of Northern Cyprus. This sovereign entity lacks international recognition and remains isolated from the rest of Europe.

Turkey has previously stated it will boycott the Cyprus presidency and warned it may freeze relations with the EU, and even formally annex the north.

Earlier this week, hundreds of Turkish Cypriots staged a protest on the streets of Brussels against the upcoming presidency, saying the rights of the island?s Turkish minority were being ignored.

A better Europe

Yet Cyprus? political class remains undeterred by the difficulties. In May, Deputy Minister to the President for European Affairs Andreas Mavroyiannis said: ?The Cyprus issue and the Cyprus presidency of the EU are two parallel and independent processes, and we will not accept to have a second-class presidency due to our national problem and to the stance of Turkey towards the Council of the EU.?

The country?s main objective for the presidency is ?to work towards a Better Europe, a Union more relevant to its citizens and the world.? This agenda will be pursued against two themes of ?solidarity and social cohesion? and ?effectiveness and growth?.

Mavroyiannis conceded the subjects that Cyprus will be able to address have been ?more or less predetermined? since these are inherited from the previous presidencies and from the EU?s 2012 work programme.

However, there is still room to champion a few signature issues. Chief among these for Cyprus include negotiations of the new Multi-annual Financial Framework for 2014-20, which is a mechanism for keeping EU spending predictable and subject to strict budget discipline.

And for all Turkey?s grandstanding, Mavroyiannis also had an olive branch to extend: ?The reinforcement of Turkey?s accession prospect is of critical importance, and the presidency will focus on advancing this prospect,? he said.

Singapore church defends pastor, officers over fraud

AFP - A Singapore-based Christian church rocked by a massive financial scandal has stood by its founder and four aides who could face life imprisonment for allegedly misusing the congregation's funds.

Pastor Kong Hee, 47, founder of the City Harvest Church, and four officers were charged on Wednesday with varying degrees of involvement in a scheme to siphon off Sg$24 million ($19 million) to finance his wife's singing career.

In addition, more than Sg$26 million in church money was allegedly misappropriated to cover up the original diversion.

In a statement posted on its website late Thursday, the church denied any wrongdoing by the five, who are out on bail while on trial.

"It has been suggested that the church has been cheated of Sg$50 million. This is not accurate," executive pastor Aries Zulkarnain said in the statement.

"The Sg$24 million, which went to investment bonds, was returned to the church in full, with interest. We didn't lose the $24 million, nor did we lose 'another $26.6m' as alleged," he added.

"The church did not lose any funds in the relevant transactions, and no personal profit was gained by the individuals concerned."

Zulkarnian, a co-founder of the group, said Kong will continue preaching at the church, which has a membership of more than 30,000 and assets estimated at more than Sg$100 million in 2009.

Meanwhile, the accused pastor's wife, who goes by the name Sun Ho in the entertainment industry, has been widely ridiculed by Singaporeans who dismissed her claims of using music to foster the church's evangelical mission.

Now believed to be in her early 40s, she based herself in Hollywood while trying to break into the international music industry.

Videos of Sun Ho's song and dance numbers on YouTube -- many featuring her in skimpy clothing and dancing provocatively -- have become overnight sensations since the scandal broke, but for the wrong reasons.

Ho's previously obscure music video "Mr Bill", where she plays an Asian wife who calls herself a "geisha" and sings about killing her black husband, had been viewed more than 150,000 times by Friday morning.

"Isn't 'thou shalt not kill' one of the ten commandments?" a YouTube viewer commented on the video-sharing site.

Kong and his wife became minor celebrities in Singapore after launching the church, which has affiliates in neighbouring Malaysia and other countries.

The church is known for services that resemble pop concerts.

Singapore authorities have cracked down on heads of charities found to have been involved in irregularities.

In 2009, a prominent Buddhist monk was jailed for six months for misappropriating hospital funds and lying about it to authorities.

Ex-Sands exec alleges prostitution in Macau sites

(AP) ? The fired former chief executive of Las Vegas Sands Corp.'s Macau casinos alleges in court documents revealed Thursday that billionaire Sheldon Adelson personally approved of prostitution and knew of other improper activity at his company's properties in the Chinese enclave.

Brad Brian, an attorney representing Las Vegas Sands Corp. and Sands China Ltd., called the allegations false and "scurrilous" and claimed they had been included in the civil lawsuit brought by former Sands executive Steven Jacobs only to sensationalize the case.

Adelson, a billionaire philanthropist and casino mogul who owns the Venetian and Palazzo casinos on the Las Vegas Strip, also is a prominent political donor who gave $10 million to a super PAC backing then-presidential hopeful Newt Gingrich and more recently has supported Mitt Romney's bid for the White House.

Jacobs was fired in July 2010 from his role overseeing the Macau properties. He sued the companies and Adelson three months later.

In the lawsuit, he accuses the company and Adelson of breach of contract and of pushing him into illegal activity in Macau, a former Portugese colony near Hong Kong where Sands has established a strong business presence. The company owns the Venetian Macao and Sands Macao casino resorts, the Plaza Macao hotel, restaurant and shopping complex and the newly opened Sands Cotai Central resort with three hotels and two casinos.

In documents revealed Thursday ? including a sworn seven-page declaration that Jacobs submitted along with a summary from his attorneys of problems obtaining documents from Sands ? Jacobs describes an effort he launched after arriving in Macau in May 2009 to rid the casino floor of "loan sharks and prostitution."

"This project was met with concern as (company) senior executives informed me that the prior prostitution strategy had been personally approved by Adelson," Jacobs said in the documents.

In his court filing, Jacobs alleges other documents that haven't been turned over include records of misuse of "blue card' work permits and the hiring of illegal workers in Macau; emails and records of Adelson controlling a "Chairman's Club" allowing favored members, including known or suspected organized crime figures, exclusive access to Sands China's most luxurious accommodations; and email requests from Adelson to a Macau lawmaker who Jacobs said was hired as outside counsel after Jacobs was fired.

Las Vegas Sands spokesman Ron Reese said in a statement that allegations of misconduct and wrongdoing by Jacobs against the company and senior managers are baseless.

"Mr. Adelson has always objected to and maintained a strong policy against prostitution on our properties and any accusation to the contrary represents a blatant and reprehensible personal attack on Mr. Adelson's character," the statement said.

In court, Brian told Clark County District Court Judge Elizabeth Gonzalez that if Jacobs "truly believed that Mr. Adelson had approved prostitution, he would have resigned." Instead, Brian said, Jacobs was fired.

Nevada Gaming Control Board Chairman Mark Lipparelli wouldn't say Thursday whether the state regulatory agency is investigating Jacobs' claims against his former employer. The agency doesn't comment on probes, he said.

Jacobs' lawsuit has drawn interest from U.S. Justice Department and Securities and Exchange Commission investigator for possible violation of the Foreign Corrupt Practices Act, according to a filing with the SEC. The law bars American companies from paying foreign officials to "affect or influence any act or decision" for commercial benefit. No charges have been filed.

The company and Adelson deny wrongdoing. They say the investigations stem from allegations by Jacobs. Sands also alleges that Jacobs stole confidential documents that might contain trade secrets.

The case once was scheduled for trial this month, but evidentiary hearings have been postponed amid clashes between the two sides about trading information and documents.

The judge scheduled a July 13 hearing on possible sanctions against the company and its lawyers for failure to disclose to the other side and to her that some documents sought by Jacobs' legal team had been brought from Macau to the U.S. more than a year ago.

With options like fresh mint with lemon lavender shortbread cookie dough and coffee ice cream with spicy chocolate cookie dough, you can't go wrong.

  • Coolhaus

What is Coolhaus? An ice cream sandwich deconstructed into a cookie roof and floor with ice cream walls. In other words, it's amazing. Especially the potato chip and butterscotch cookie with molten chocolate cake ice cream. Mmmmmmmm.

  • Chinatown Ice Cream Factory

A Chinatown landmark, CICF takes traditional ice cream and puts a Chinese twist on it -- in this case red bean, black sesame, almond cookie and zen butter to name a few.

  • Love Gelato

  • Ample Hills Creamery

Local and artisanal but without taking itself too seriously, Ample Hills Creamery churns out some of Brooklyn's fnest ice cream. Stop by the Prospect heights shop for a scoop of Ooey Gooey Butter Cake or a Salted Crack Caramel cone. Delish!

  • Van Leeuwen Ice Cream

These guys take their ingredients very very seriously, and the result is magic in your mouth. Van Leeuwen Ice Cream is made solely of fresh milk and cream, cane sugar, egg yolks and "the finest flavors from small producers locally and around the world" (think pistachios from Mount Etna in Sicily, Piedmont hazelnuts, hand crushed quills of ceylon cinnamon from Sri Lanka)

  • Big Gay Ice Cream

It's the toppings that differentiate Big Gay Ice Cream (well that and the name). His schtick is simple: traditional soft serve ice cream with totally-out-there, unexpected toppings and kitschy names. We recommend the Bea Arthur (vanilla with dulce de leche and crushed 'nilla wafers) or the Mermaid (vanilla with key lime curd, crushed graham crackers, whipped cream).

  • Il Laboratorio del Gelato

With over 200 flavors to choose from, Il Laboratorio del Gelato is truly a lab. To ensure the highest quality ice cream and sorbet, each is made in small batches of the finest ingredients, sourced from all over the world.

Case in point: The Almond Joyful Sundae -- small batch coconut ice cream topped with hot fudge, Farmacy's own coconut almond crumbles and fresh whipped cream.

  • L'Arte del Gelato

  • Blue Marble Ice Cream

Blue Marble serves up award-winning ice cream with a conscience. With shops in Prospect Heights, Boerum Hill and Rwanda (for real!), they keep the focus on local, simple ingredients, always avoiding sugary add-ins, preservatives, corn syrup, and artificial flavors or coloring.

Renting Out Vacation Condos

Wouldn't it be nice to own your own vacation home where you can escape for a weekend and enjoy the mountains or the beach without having to research hotels and condos? Many dream of owning their own second home to be used as a vacation place, but few actually take steps to realize their dream. Depending on your location, owning a vacation home nearby is not only convenient for frequent unplanned vacations, but it can also be a means of income.

Location, Location, Location

Where do you enjoy vacationing? If you live just hours from the mountains or the beach, either of these options may provide a wonderful place to purchase a second home. Focusing on condos will probably provide many additional amenities like a community swimming pool, fitness area, tennis courts, beach access, or cross country ski trails. If you're a big city person, a condo in the city provides easy access to restaurants, stores, and the local bus system, which can provide easy transportation to places of interest.

Tiananmen anniversary keeps Chinese censors on edge (+video)

Beijing still shows extreme sensitivity to the 1989 crackdown in Tiananmen Square. China's version of Twitter is censoring everything from today's date to the word 'candle.'

?For the 23rd year in a row, the Chinese authorities today continued their efforts to impose collective amnesia about the Tiananmen Square massacre on June 4, 1989, seeking to stamp out any public reference to the event.

For Beijing, the date, ?6.4? as it has become known in China, is one not to be commemorated, but rather obliterated from the calendar. The government has always refused to discuss what happened when soldiers were ordered to disperse pro-democracy demonstrators on Tiananmen Square. Officially, they killed only about 200 protesters; but activists? estimates range from several hundreds to several thousands.

The official blanket denial of the date has worked, to a large extent: Few ordinary Chinese citizens under the age of 30 are aware of the Tiananmen demonstrations or their tragic end. But censors remain determined to foil any attempt by people who do know what happened to say anything about it. Anything at all.

Censors at Sina Weibo, the popular Twitter-like social media platform, were working overtime to block searches for ? or references to ? ?6.4? or other obvious signifiers such as ?tank,? ?crush,? ?never forget,? and ?square.?

?535? was a forbidden term, too, because Internet users have taken to referring to May 35,? instead of June 4. Classically minded censors wouldn?t let you post anything with VIIIIXVIIV in it either, in case readers familiar with Roman numerals could decipher 89.6.4. And by late afternoon, even the word ?today? had been banned.

Over the weekend, Sina Weibo disabled its candle icon, which users might find an appropriate symbol with which to commemorate the Tiananmen dead. An explanation from the website said the icon was ?currently being optimized? but nobody believed it. Before long there were 200,000 posts on Sina referring to ?Weibo? and ?candle? and the next thing anyone knew, the word ?candle? itself became a banned search term.

As if to highlight the absurdity of how hard Sina Weibo censors were working, an impeccable and seemingly?irrelevant source of information, the Shanghai Stock Exchange, suddenly turned subversive. By an extraordinary coincidence (or maybe not), the Shanghai Composite Index, which measures the market?s daily movement, fell on Monday by 64.89 points, in an uncensorable reminder of the date of the massacre.

Though the official campaign to airbrush the Tiananmen Square events out of Chinese history has been successful with most citizens, there are some, of course, who can never forget because their own children died.

One such father, Ya Weilin, apparently exhausted and driven to despair by his failure after nearly a quarter of a century to persuade the government to account for his son?s death, hanged himself 10 days ago. Tiananmen is still claiming victims; 23 years ago they were felled by bullets. Today they succumb to silence.

Event: "Reset: post-consumer gamer culture" (June 21 to July 28 ...

"Pong?was the first. Its ability to entertain people for hours on end with no visible?narrative or plot line was the feature that distinguished it from other forms of media, namely music and movies. It was the setting that counted here, and the setting alone. Of course,?Pong?s was only a preview of what was to come; a multi-billion dollar video game industry that has both affected and influenced pop culture in ways that most of us may not even realize.

Are you a fan of the?Resident Evil?films? 1980s hip hop? What about?Lara Croft: Tomb Raider,?Grandma?s Boy?or?Tron? Can you hum the?Super Mario Bros.?theme song on command? I?ve never played the game in my life, and I can do it. The list goes on and on. Even though the influence of video games can be easily seen in mainstream pop culture, what about considering how gamer culture and the rise of digital media have influenced our attitudes toward art? Using video games as the medium is one of the more unexplored avenues of the influence of gamer culture, but it?s exactly what an upcoming Winnipeg exhibition is aiming to do.

Reset: post-consumer gamer culture, at the Platform Centre for Photographic and Digital Arts starting June 21, explores the connection between video games and art, and how gamer culture has infiltrated and influenced society around us. Through new media work in print, projections, and hand held interactive forms,?Reset?explores the ways in which video games have evolved from mere pixels depicting the simplest of scenes to an international industry with countless brands, ?worlds,? and affiliations.

The artists whose work is featured have embraced the technology, culture and aesthetics of gaming as a medium. Their work also highlights the history of gaming technology through the devices that the pieces are viewed on. Some use the aesthetics of the games to create digital drawings and short animated loops; some use the technology as a filmmaking tool (this is known as ?machinima?); others create their own interactive software applications and modify game controllers or interfaces.

Among the artists whose work will be included in the exhibition are native Winnipegger Clint Enns, a filmmaker whose work centers around creating something new out of broken and/or outdated technologies, and Canadian Myfanwy Ashmore, known for her software artwork. Also featured is the work of Dr. Ian Bogost, an award winning designer and media philosopher whose games cover social and political issues and have been played by millions of people internationally. Max Capacity, a media and technology artist from Santa Cruz, California is also featured, his glitch work being derived from a variety of dead media like vintage video games, old and malfunctioning VHS machines and bent electronics.

?Party Time! Hexcellent!? is the name used by Rachel Weil, an artist who creates interactive, glitchable visual art for the Nintendo Entertainment System. And finally, Haydi Roket is an independent graphic designer and media artist from Istanbul whose work is preoccupied with the aesthetics of 8-bit computer and gaming technology; vintage video game consoles and computers are the source of his pieces.

While the sheer variety here would make for more than enough talent for one exhibition, the entire lot is curated by mrghosty (also known as Skot Deeming), a media artist as well as a curator whose own work seeks to challenge consumer models of play associated with video games and gaming culture.

Many various industries are now moving in digital directions. The predictable result is the convergence and interconnection of film, music, fashion, video games and now fine art. With such a unique form of art created by such considerable talents on display in Winnipeg, this exhibition isn?t one to miss.

Reset: post-consumer gamer culture is on at the Platform Centre for Photographic and Digital Arts from June 21 to July 28. The Opening Reception and Curator?s Tour is on Thursday, June 21 at 7 p.m."

(AP) ? Unlike a lot of movies shot in Louisiana, "Beasts of the Southern Wild" couldn't have been made anywhere else.

Louisiana natives and the state's bayous and marshes tell the fictional tale of a 6-year-old girl named Hushpuppy and her struggle for survival in the southern Delta with her ailing father as a storm approaches.

The film is gripping audiences around the globe for its portrayal of the spirit and resiliency of the people of south Louisiana through the fictional tale of a motherless young girl trying to hold on to her place in the world ? that place being a small, tightly-knit shantytown on the bayou just beyond the government's levee protection system. Her world is filled with wild animals, both real and imaginary.

"Beasts of the Southern Wild" was shot in southern Terrebonne Parish and Isle de Jean Charles, a real bayou community that is gradually eroding away because it has no levees to protect it. But the film's makers say it isn't a social issue movie.

"It's really about what it's like to lose the thing that made you," said director Benh Zeitlin, a native New Yorker and one of only a handful of people affiliated with "Beasts of the Southern Wild" not from Louisiana. "The film is inspired by culture and events in Louisiana but we wanted to elevate it to make it universal. It's about where you come from. Your land is your parent, and there are all these emotional connections that stem from where you come from."

"Beasts of the Southern Wild" won the grand jury prize in the U.S. dramatic competition at the Sundance Independent Film Festival as well as its cinematography award. At France's Cannes Film Festival in May, where many U.S. films were shut out, Louisiana's indie darling won the Camera d'Or for best first film.

Zeitlin said the geography of the film didn't even exist until he found the community of Pointe-aux-Chenes and just beyond it Isle de Jean Charles ? the last inhabited speck of land at the end of a winding highway south of Houma, La.

"I started driving to the end of all these south Louisiana roads, and when I drove to the end of this one, I knew I had found the place that would be the setting of this film," Zeitlin said.

Finding the right actors to play the leads took more searching. Producers went on a months-long casting quest for the child who would play Hushpuppy. It included auditions of nearly 4,000 girls in countless schools and churches across south Louisiana.

They found 5-year-old Quvenzhane (Kuh-VAHN-zuh-nay) Wallis of Houma, La., who was 6 at the time the movie was filmed in 2010. Nazie (NAY-zee), as she likes to be called, had a quality producers couldn't resist: "It's her eyes. There's something in her eyes that gives her poise, maturity, depth and wisdom beyond her years," Zeitlin said.

Now 8, Nazie walked the red carpet Monday evening at the New Orleans premiere of "Beasts" in a bright blue dress, silver slippers and a big smile as she talked to reporters and posed for pictures.

Her hair straightened and bangs pulled in a tight braid above her forehead, Nazie bore little resemblance to the pants-wearing bushy-haired wild child she portrays in "Beasts."

Also walking the carpet was New Orleans native Dwight Henry, who was hand-picked for the role of Hushpuppy's father, Wink, after producers discovered him in the Buttermilk Drop Bakery he runs in New Orleans.

"We went there every morning for grits and doughnuts, and we got to know him," Zeitlin said. "He talked about his life, things he had been through, and parts of his life got written into the film. At some point, we realized he was the one we wanted for this film."

Henry turned them down three times.

"I'm glad I did it, but at the time, I was reluctant to do it, to sacrifice my business that I had worked so hard to open for a chance at a possible movie career," said Henry.

Now Henry and Nazie say they are both looking forward to more film work. Henry has partners helping out at the bakery while he works on his film career, including a part in "Twelve Years a Slave," about a New Yorker who was captured in 1841 and enslaved in Louisiana. The film, directed by Steve McQueen, will also star actors Brad Pitt and Michael Fassbender.

Zeitlin said his affinity for New Orleans grew out of events surrounding Hurricane Katrina and through a short film he had been working on at the time the hurricane struck called "Glory at Sea," which all led to the making of "Beasts of the Southern Wild."

The Fox Searchlight film opens in select theaters, starting with showings in Los Angeles and New York on Wednesday and then New Orleans-area theaters July 4.

Understanding Senior Care and Elder Law

Senior care and elder law are closely related industries that seek to provide care and protection for one of our most vulnerable populations -- senior citizens. But while distinctly different, senior care and elder law often intersect when it comes to the needs of our aging family members. Understanding what each is and how it can help your elder loved one can significantly improve the lifestyle of your entire family.

Senior care is a healthcare field dedicated to providing basic assistance and medical care for seniors. The field broadly refers to three primary types of care:

In Home Care -- The type of care that allows for the highest level of independence, in home care is provided by skilled professionals in the home of the elderly person. This can include full time, part time or relief care services.

Assisted Living -- Assisted living generally refers to a community designed, built and managed in such a way as to provide long term support to seniors. Members of assisted living communities often live in their own apartment or condominium and have access to a variety of senior care services.

Nursing Care -- Nursing care is the most intensive level of care and offers full medical services including medication management. Nursing care residents live in a specialized facility staffed by nurses and their aides, and in some cases psychiatrists and medical doctors are part of the regular staff.

Elder law is a relatively new field of law that deals exclusively with legal issues related to senior citizens. This includes estate planning, wills and trusts, laws dealing with insurance and medical care, and laws that protect the elderly from neglect or abuse.

Senior care and elder law intersect in the following 2 ways:

1.) Life Decisions

There are complicated laws surrounding legal issues like wills, long term care and estate planning. These laws require legal counsel even in the most ideal situations, but what often happens is that families begin asking about these laws only after a crisis has occurred.

For instance, in the case of an elderly family member suddenly taking ill and being unable to make their own rational decisions, legal intervention is often required in order for long term care to proceed. And in cases of dementia where family members want to provide care at home; there are legal responsibilities that must be undertaken when doing so.

The best solution to these problems is to create a plan before crisis occurs.

2.) Protection from Abuse

It's unfortunate that we would need them, but there are laws in place that protect seniors from neglect or abuse. In many cases the punishment for violating these laws are severe and come with hefty prison sentences. Family members that suspect their loved ones are being mistreated can take action by invoking elder law. Authorities are required to rigorously investigate any such claims.

Senior care is ultimately governed by elder law, so developing an understanding of the two is critical to providing a lifetime of support for an elderly family member. To learn more, call a professional in home care giver or senior care provider for an immediate consultation.

Glucose deprivation activates feedback loop that kills cancer cells, study shows

ScienceDaily (June 26, 2012) ? Compared to normal cells, cancer cells have a prodigious appetite for glucose, the result of a shift in cell metabolism known as aerobic glycolysis or the "Warburg effect." Researchers focusing on this effect as a possible target for cancer therapies have examined how biochemical signals present in cancer cells regulate the altered metabolic state.

Now, in a unique study, a UCLA research team led by Thomas Graeber, a professor of molecular and medical pharmacology, has investigated the reverse aspect: how the metabolism of glucose affects the biochemical signals present in cancer cells.

In research published June 26 in the journal Molecular Systems Biology, Graeber and his colleagues demonstrate that glucose starvation -- that is, depriving cancer cells of glucose -- activates a metabolic and signaling amplification loop that leads to cancer cell death as a result of the toxic accumulation of reactive oxygen species, the cell-damaging molecules and ions targeted by antioxidants like vitamin C.

The research, which involved UCLA scientists from the Crump Institute for Molecular Imaging, the Institute for Molecular Medicine, the California NanoSystems Institute, the Jonsson Comprehensive Cancer Center, the Eli and Edythe Broad Center of Regenerative Medicine and Stem Cell Research, and the Department of Pathology and Laboratory Medicine, demonstrates the power of systems biology in uncovering relationships between metabolism and signaling at the network level.

"Most strikingly, our discovery that glucose withdrawal causes both cell death and increased tyrosine phosphorylation is intriguing because increased tyrosine kinase signaling is normally associated with cell growth," said Nicholas A. Graham, a senior postdoctoral scholar in Graeber's lab who helped design the project.

To explain the seemingly contradictory result that glucose deprivation reduced viability and at the same time increased signaling, the authors used an unbiased systems-biology approach that included phospho-tyrosine mass spectrometry and other biochemical profiling techniques.

Assessing the "crosstalk" between metabolism and signaling, they discovered that the glucose deprivation activates a positive feedback loop whereby the withdrawal of glucose induces increased levels of reactive oxygen species, which in turn inhibit negative regulators of tyrosine signaling. The resulting supra-physiological levels of tyrosine phosphorylation then generate additional reactive oxygen species.

"Because cancer cells live on the edge of what is metabolically feasible, this amplifying cycle of oxidative stress ultimately overwhelms and kills the cancer cell," Graeber explained. "These findings illustrate the delicate balance that exists between metabolism and signaling in the maintenance of cancer cell homeostasis."

In addition, the authors showed the possibility of exploiting this positive feedback loop for therapeutic intervention. Combining short-term glucose deprivation with an inhibitor of tyrosine phosphatases, they demonstrated synergistic cell death in a cancer cell line.

"Understanding the links between metabolism and signaling will empower new therapeutic approaches toward inducing this metabolic catastrophe," Graham said. "This study provides a framework for rational design of combinatorial therapeutics targeting both metabolism and signaling in cancer."

The findings by Graeber and his colleagues add to the emerging concept of systems integration between oncogenic signaling networks and the metabolism of malignant tumors. The work lays a foundation for future studies delineating how signaling and metabolism are linked, with the ultimate goal of refining therapeutic strategies targeting cancer metabolism.

The research team also included collaborators from the department of neurology and the human oncology and pathogenesis program at Memorial Sloan-Kettering Cancer Center and the department of pharmacology at Weill-Cornell Medical College.

The research was funded by the National Institutes of Health, UCLA's Jonsson Comprehensive Cancer Center, and the California Institute of Technology-University of California, Los Angeles, Joint Center for Translational Medicine.
